The serine/threonine-protein kinase Receptor Interacting Protein Kinase 1 (RIPK1) is required for tumor necrosis factor receptor 1 (TNFR1)-driven necrotic cell-death (necroptosis). When TNFR1 is stimulated by TNF or Fas, it interacts with adaptor proteins (FADD and TRADD), which bridge the interaction between TNFR1 and RIPK1. Activated RIPK1 then associates with RIPK3 to form a necroptosis-inducing complex. RIPKs are also responsive to pathogen recognition receptors (PRRs), inflammation and DNA damage. Necroptosis serves as an alternative to apoptosis and is implicated in the pathogenesis of a variety of human diseases such as brain injury, immune disorders and cancer.
